I don't know if Dean knows that Castiel let Sam out, but he does know that for a time, Castiel was in on the Angels' big plan to start the Apocalypse... one way or another, Castiel was convinced that human lives were nothing but sorrow, and the Apocalypse was the Angels' solution. Dean convinced him otherwise -- but now Cas has to wrestle with his own doubts about diverging from the will of heaven -- something he's never done before and that evidently makes him feel frightened, angry, a little guilty, and consequently a little quick to blame his allies that he went along with them.
